A specimen that falls outside the normal range for any of these markers gets flagged as dilute, substituted, adulterated, or invalid, and that finding is treated as seriously as a positive result in most court and probation settings.
This matters because attempting to beat a drug test isn’t just a testing problem, it’s a legal one. In a probation, parole, or family court context, a flagged specimen can trigger the same consequences as a confirmed positive, sometimes with less room to argue the result.
What specimen validity testing actually checks
Every legitimate testing program runs validity checks alongside the drug panel itself, not as an afterthought. The core markers are:
Temperature. A urine specimen has to register within a narrow window (typically 90 to 100 degrees Fahrenheit) within minutes of collection. Anything outside that range is a strong signal the sample didn’t come directly from the donor’s body.
Creatinine and specific gravity. These measure how concentrated the urine is. Creatinine that’s too low, combined with a specific gravity close to that of water, points to either excessive fluid loading or outright substitution with a non-urine liquid.
pH. Human urine sits in a fairly predictable acidic-to-neutral range. A pH that’s unusually high or low often means a chemical adulterant, like bleach or a commercial “detox” product, was added to the sample.
Oxidizing adulterants. Tests specifically screen for nitrite, chromium, halogens, and other oxidizing agents that are designed to break down drug metabolites in the sample before it reaches the lab.
When a specimen fails one or more of these checks, most programs classify it in one of four ways: dilute (too much fluid, not necessarily intentional), substituted (not consistent with human urine at all), adulterated (a foreign substance was added), or invalid (results are inconsistent and can’t be interpreted). Each classification carries a different procedural response, and that distinction is often what gets litigated when a result is challenged.
Why the distinction between dilute and adulterated matters
A dilute specimen isn’t automatically treated as an attempt to cheat. Someone who drinks a lot of water before a scheduled test, intentionally or not, can produce a dilute sample without any chemical interference. Most programs handle a first dilute result with a same-day or next-day recollection rather than a violation.
An adulterated or substituted result is a different category entirely. Because it points to deliberate tampering rather than hydration, it’s typically treated procedurally the same as a confirmed positive, including in many probation and drug court settings. This is the edge case worth understanding: a donor who genuinely didn’t try to cheat can still land in the more serious category if their specimen happens to fail validity testing for an unrelated reason, which is why documented, defensible collection procedures matter so much.
What the guidelines actually require
The federal Mandatory Guidelines for Workplace Drug Testing Programs, published by SAMHSA, lay out the specific cutoff values labs use for creatinine, specific gravity, pH, and oxidant screening, along with the required confirmatory steps before a specimen can officially be reported as adulterated or substituted. Many state and county court systems that don’t operate under federal jurisdiction still model their own specimen validity protocols on these same cutoffs, because they represent the most scientifically vetted standard available.
That’s also why chain of custody and validity testing tend to travel together in any program built to survive a legal challenge. A lab can run a technically accurate validity test, but if the collection, handling, and documentation around that specimen aren’t equally rigorous, the result becomes easier to dispute. Programs that source their drug testing supplies with validity testing built into the collection process, rather than added on afterward, tend to generate fewer disputed results down the line.
